DM-4103 was dissolved in DMSO and administered to male rats at doses of 100 and 500 mg/kg by single subcutaneous administration, since DM-4103 is insoluble in water and is re-crystallized when mixed with serum or injected intravenously. DM-4103 did not show any notable toxicity at 100 mg/kg (Cmax: 54 g/mL, AUC24h: 743 g•h/mL) or at 500 mg/kg (Cmax: 26 g/mL, AUC24h: 319 g•h/mL). The reversed exposure observed in this study was due to the reversed absorption amount, higher at 100 mg/kg and lower at 500 mg/kg, due to re-crystallization of DM-4103 at the injection site. DM-4107 was also dissolved in DMSO and subcutaneously administered to male rats at single doses of 100 and 500 mg/kg. No toxicity was noted and the serum level of DM-4107 increased with dose elevation, with a Cmax 310 g/mL and an AUC24h of 3906 µg•h/mL.
