4.2.2 Material and methods
4.2.2.1 Construction and analyses of a single reservoir diffusion gradient
plate

A physical model was constructed of Plexiglas (Fig. 4.2.l.A). Agarose (1.5%)
was used to prepare gels, 38 mm long, 16 mm wide, and 5 mm thick. Diffusion from a
single reservoir (dimensions 0.32 x 16 mm x 0.60 mm, located below the gel, 0.32 mm
from one end), filled with a test compound, resulted in the development of a concentration
gradient at the surface of the gel. The content of the reservoir was replaced regularly to
ensure maintenance of a constant concentration in the reservoir. A constant flow of sterile
water over the surface of the gel (Q=650 mm h-1, flowing from right to left in Fig.
4.2.l.A, removed the test compound as it diffused out of the gel. A 50 mm long #1
microscope coverslip, mounted on the Plexiglas using silicon adhesive (General Electric
RTV), was used to cover the gel. This enabled microscopic examination of biofilm
development on the gel surface with an extra long-working-distance objective (Nikon
Corporation, Chiyoda-ku, Tokyo). Alternatively, a water immersion lens (Nikon Corp)
was used after the coverslip had been removed.

peristaltic pump. The small size of this model (dimensions of the gel were 38 by 16 rnm by 5 mm thick, distance between the gel and coverslip was 0.5 mm) enabled direct microscopic observations of biofilm development on the surface of the gel. The number of reservoirs and solute concentration in the reservoirs were altered in the modified gradient plate (not shown) to manipulate the diffusion profile in the gel. (8) Flow and concentration boundary conditions, and the overall problem size in the LTG simulation.

Ayudaría mucho saber de qué procedimiento concreto se trata. Al parecer no es electroforesisi, de modo que reservoir bien podría ser traducido como reservorio. Mi propuesta: placa de un solo pozo/pocillo/reservorio para gradiente de difusión (en gel de agarasa, por lo que parece)

WHY IS DIFFUSION AN IMPORTANT PROCESS IN BIOFILMS?
When microorganisms are grown in planktonic culture, diffusion is usually of little consequence. There are two reasons for this. The first reason is that planktonic cultures are generally agitated, and the resulting fluid flow transports solutes rapidly, resulting in a well-mixed system. Transport that occurs as a solute is carried by the bulk flow of a fluid (convection) is generally much faster than the transport resulting from random molecular motion (diffusion). Of course, there is no net convective flow of fluid into or out of the microbial cell. At some point close to the cell, diffusion becomes critical for moving the solute toward or away from the cell surface. The reason that diffusion does not limit this step is that the diffusion distance is small and diffusion is rapid over such short distances.
