Source text - English Tylosin is a macrolide antibiotic isolated from Streptomyces fradiae. Chemically and in mechanism of action it is similar to other macrolide antibiotics, such as erythromycin. Source text - English Ivermectin is obtained by chemical modification of avermectin B1a, a macrocyclic lactone obtained from the actinomycete Streptomyces avermitilis NRRL8165. The structural and molecular formulae of ivermectins are shown below.
Ivermectin is used widely in veterinary medicine as a broad-spectrum antiparasitic drug, particularly against nematodes and arthropods in cattle, horses, pigs and sheep. It is also used in humans for the treatment of onchocerciasis, a disease of Africa and parts of tropical America caused by the microfilarium Onchocerca volvulus which is transmitted by various species of black fly and which results in inflammation of the skin and in some cases blindness (‘river blindness’).
In general, parasites are controlled by a single treatment with ivermectin, but year-round parasite control programmes may involve 3 or 4 treatments per year.
This Application relates to the use of XXX in horses and donkeys.
XXX Horse Paste contains Ivermectin (1.87% w/w), hydrogenated castor oil, hydroxypropyl cellulose, titanium dioxide and propylene glycol. All constituents of Ecomectin Horse Paste are of European Pharmacopoeia grade. All excipients in this formulation are commonly used in veterinary medicine. Further considerations of the formulation are presented below in section 4.4.
As stated above, ivermectin is one of a group of compounds called avermectins and is a highly potent, broad-spectrum antiparasitic drug active against nematodes and arthropods. The avermectins are macrocyclic lactones obtained as fermentation products of an actinomycete, Streptomyces avermitilis. It has been shown that, in target organisms, the effect of ivermectin is mediated through a specific, high affinity (ca. 10-10M) binding site, which results in an increase in membrane permeability to chloride ions, this being independent of GABA-mediated chloride ion channels. Although GABA-gated chloride channels do also interact with ivermectin, this occurs only at concentrations around a thousand times greater (i.e. 10-7M), so this is probably not relevant to the clinical use of ivermectin. Indeed, it may explain why the drug is so well tolerated by host vertebrate species at antiparasitic dosage levels.
The result of ivermectin-induced increase in membrane permeability to chloride ions is that post-synaptic transmission of nerve impulses is blocked so that excitatory stimuli are not received by motor neurones in nematodes, and neither excitatory nor inhibitory stimuli are received by arthropod muscle cells. These effects of ivermectin are irreversible and prolonged. The end result is paralysis and death of the parasite.
Ivermectin was tested in vitro for genotoxic activity in microbial (Ames test) (See Tabulated Study Report No. 21) and mammalian cell (mouse lymphoma) (See Tabulated Study Report No. 22) mutagenesis assays as well as in human fibroblasts for DNA damage as measured by unscheduled DNA synthesis (See Tabulated Study Report No. 23). In all instances, ivermectin had no genotoxic effect (Ref. 11). There is a report that ivermectin induced sister chromatid exchanges (SCE) in human lymphocytes in vitro at concentrations of 0.3 mcg/ml and above. No increase in micronuclei was observed. There was no effect on SCE at 0.15 mcg/ml (Refs. 16, 17; Tabulated Study Report No. 24). These data do not indicate any genotoxic hazard to humans (or animals). It can be calculated from the data discussed in Section 2.2 above that the Cmax for ivermectin in humans at the therapeutic dose is approximately 0.02 mcg/ml, so the no-effect concentration in the human lymphocyte study is 7.5 times the human Cmax. Moreover, in the absence of a positive result in at least one other standard genotoxicity test, a positive in the SCE test is normally regarded as of little or no biological significance.

In tutti i casi, l’ivermectina non ha manifestato alcun effetto genotossico (Ref. 11). Vi è una segnalazione che l’ivermectina possa indurre, in vitro, scambi tra cromatidi fratelli (SCE) in linfociti umani a concentrazioni pari o superiori a 0,3 mcg/ml. Non è stato osservato alcun aumento nei micronuclei. Non vi era alcun effetto su SCE alla concentrazione di 0,15 mcg/ml (Ref. 16, 17; Rapporto sullo studio tabulato N.24). Questi dati non indicano alcun rischio di genotossicità per gli esseri umani (o per gli animali). Può essere calcolato, dai dati discussi nel precedente Paragrafo 2.2, che la Cmax per l’ivermectina alla dose terapeutica sia, nell’uomo, approssimativamente di 0,02 mcg/ml, quindi, la concentrazione priva di effetto nello studio sui linfociti umani supera di 7,5 volte la Cmax nell’uomo. Hinter Lachen und Gewimmel in den glitzernden Wassertexturen des XXXs® steht die Philosophie des Designteams von YYY. Ein XXX® besteht aus mehreren einzelnen Play Events, die verschiedene Wassererlebnisse intelligent kombinieren. Jedes für sich hat eine eigene Dynamik und passt zu einer bestimmten Altersgruppe. Wir planen XXXs® in allen erdenklichen Größen zwischen 30m² und über 700m². Das Kernareal konzipieren wir als Family Bay – dem Anziehungspunkt für alle Generationen. XXXs® laden alle Menschen gleichermaßen mit ihren unterschiedlichen Fähigkeiten ein, die elementare Begegnung mit den schönsten Aspekten des Wassers zu genießen. Als einziges Freizeitareal bieten XXXs® Menschen mit körperlichen und geistigen Behinderungen ganz genau die gleichen Möglichkeiten wie allen anderen. XXXs® sind weit mehr als nur «barrierefreie» Freizeitareale. Sie definieren ein integratives Erlebniskonzept, das weit über die amerikanischen ADA Anforderungen hinaus weist. Toddler Bay – die spannende Entdeckungsreise
Ab einer bestimmten Größe empfehlen wir ergänzend zur Family Bay die Toddler Bay - einen Bereich, in dem Kleinkinder auf sanfte Weise die Vielfalt des Wassers ungestört und eigenständig entdecken. Die unterschiedlichen Wassereffekte für diese Altersgruppe sind so eingestellt, dass sie auch kleinste Kinder nicht einschüchtern. Es entsteht eine behütete Umgebung, gestaltet mit lebhaften Farben und leichten Unterschieden in der Wassertemperatur. Kleine Pfützen mit sanft fließendem Wasser, mit kräuselnden und glatten Oberflächen und Lichtreflexen sprechen alle Sinne der Kleinsten an, passend zu ihren motorischen und mentalen Bedürfnissen und Fähigkeiten.
Teen Bay – Action und Teamgeist
Die Teen Bay ist ein anderer besonderer Schwerpunkt für Ihr XXX®. Geprägt von Teamwork, Gestaltungsmöglichkeiten und Überraschungselementen entspricht sie mit hoher Dynamik dem ungebrochenen Bewegungsdrang der größeren Kinder und Teenager. Hier gibt es Wasserschlachten mit richtig viel Wasser – hier werden alle patschnass. Diese Bay ist immer der wildeste Teil des gesamten Areals. Die Teen Bay ist ein dynamischer Bereich voller Energie. Hier wird genau das angeboten, was den meisten Kindern heute oft fehlt, ein geschützter Raum zum Laufen, Rennen, Schreien und Austoben. Die Teen Bay ist das zeitgemäße Angebot um diese Raumqualität wieder zur Verfügung zu stellen.

Play Events strukturieren die Bay
Ein gezielter Wasserstrahl, sprühende Nebelwände, platschende Wasserladungen von oben oder glatte, fast durchsichtige Wasservorhänge – Play Events kombinieren eine Vielfalt faszinierender Wassereffekte – interaktiv, überraschend oder in regelmäßigen Sequenzen. Ein Play Event ist ein inszeniertes Wassererlebnis. Es besteht aus mehreren aufeinander abgestimmten Komponenten. Die einzelnen Geräte und Effekte unterstützen in der Intensität, der Größe und der Dynamik besonders die motorischen Bedürfnisse und Fähigkeiten jeweils einer bestimmten Altersgruppe. Wir gestalten Erlebnisfolgen und Bewegungsrichtungen, die ein ausgelassenes und trotzdem harmonisches Miteinander im Raum ermöglichen.

Die Play Events der Family Bay sind so aufgebaut, dass sie das gemeinsame Erleben in der Familie unterstützen. Man kann entscheiden ob man nass wird und auf welche Weise, weil die Sequenzen der Wassereffekte verlässlich und regelmäßig stattfinden.
Elementares Erleben mit allen Sinnen
Ein Play Event in der Toddler Bay kann, dem Erlebnishorizont der Kleinsten entsprechend, schon aus einem pulsierenden Wasserstrahl mit glitzernden Tropfen und einer funkelnden Pfütze bestehen.
Raumqualität für ungebremste Motorik
In der Teen Bay sind die Play Events voller Überraschungen. Die Sequenzen sind nicht absehbar. Durch die Kombination einzelner Geräte ergeben sich hier Laufrichtungen für ungebremste Motorik. Hier gibt es das Miteinander beim gemeinsamen Gestalten von Intensität und Richtung der Wassereffekte und das Gegeneinander wilder Wasserschlachten.
Beispiel KKK ™
Entlang der Bewegungsachse dieses Play Event sind verschiedene Wassereffekte und -erlebnisse angeordnet. Am Beginn der Achse im Vordergrund befinden sich zwei Spinner. Durch die Drehbewegungen der Köpfe entsteht ein Wassereffekt, variierend zwischen Regendusche und Wassernebel. Dahinter steht der Wassertunnel zum Durchkrabbeln oder Durchrennen. Die beiden transluzenten Dumping Flowers am Ende des Play Event füllen sich langsam und lassen ihre volle Wasserladung, in absehbaren Intervallen auf die Köpfe der Untenstehenden platschen. Dies ist ein Beispiel aus der Family Bay und könnte das Zentrum Ihres XXXs ® sein.

Die Herstellungskosten im Vergleich zum Wasserbecken gleicher Größe sind bis zu 50% niedriger. Die Kinder bestimmen selbst, wann es losgeht. Sie finden und benutzen Activators auf dem Areal um die Wassereffekte zu starten. Das XXX® ist so konzipiert, dass es nur dann anläuft, wenn es vom Publikum aktiviert wird. Nach dem Durchlauf der von Ihnen ausgewählten Sequenzen, die vier bis sechzehn Minuten lang sind, schaltet es wieder ab – bis zur nächsten Aktivierung. So funktioniert es 24 Stunden an sieben Tagen in der Woche ganz nach Bedarf. Auf Ihre Wasserquelle kommt es an! Nutzen Sie das Wasser eines Pools oder eines Badegewässers und eine Pumpe für den nötigen Druck oder haben Sie bereits ein Wasseraufbereitungs-System? Möchten Sie Ihr XXX® an die Wasserleitung anschließen? YYY integriert das Command Center im passenden Modus in Ihr System. Das Command Center funktioniert anschließend vollautomatisch. Lo XXX® invita tutti indifferentemente, secondo le differenti capacità di ognuno, a liberare la naturale competitività con le più attraenti caratteristiche dell’acqua. XXX® è l’unica area divertimenti che offre a persone con menomazioni fisiche o mentali le stesse possibilità che hanno tutti gli altri. Toddler Bay – l’emozione della scoperta
A partire da determinate superfici, a completamento della Family Bay offriamo la Toddler Bay - un ambiente in cui i bambini piccoli possono scoprire i diversi aspetti dell’acqua in tutta tranquillità e indipendenza. I diversi giochi d’acqua per questa fascia d’età sono regolati in maniera tale che anche i bambini più piccoli non abbiano nulla da temere. Si tratta di un ambiente protetto, con allestimento in colori vivaci e lievi differenze di temperatura dell’acqua. Piccoli specchi di placida acqua corrente, dalle superfici increspate o calme e dai riflessi di luce che catturano tutti i sensi dei più piccoli, in armonia con le loro esigenze e possibilità motorie e psicologiche.
Teen Bay – Azione e spirito di squadra
La Teen Bay è un’altra punta di diamante del vostro XXX®. Caratterizzata dal lavoro di squadra, dalle possibilità di configurazione e dagli elementi di sorpresa risponde in maniera altamente dinamica all’indomito desiderio di movimento dei bambini più grandi e dei ragazzi. Contesti di gioco che compongono la baia
Un getto d’acqua mirato, effervescenti cortine di nebbia, sciabordanti tonfi in acqua dall’alto o a livello, muri d’acqua semitrasparenti – i contesti di gioco combinano una pluralità di affascinanti effetti d’acqua, in maniera interattiva, sorprendente o in sequenze ordinate. Un contesto di gioco è un’esperienza allestita in acqua. Si compone di molti elementi diversi. I singoli dispositivi ed effetti soddisfano l’intensità, l’entità e la dinamica delle esigenze e possibilità motorie specifiche per ogni gruppo d’età. Progettiamo sequenze e direzionalità che permettono una socializzazione sfrenata e tuttavia armonica.

I contesti di gioco della Family Bay sono progettati per offrire il giusto divertimento a tutta la famiglia. Si può scegliere se e in che misura bagnarsi, dato che le sequenze di effetti d’acqua si susseguono a ritmo regolare e prevedibile.

Un contesto di gioco nella Toddler Bay, in relazione al livello specifico dei più piccoli, può già comprendere un vibrante getto d’acqua con goccioline luccicanti e uno sfavillante specchio d’acqua.
Qualità spaziale per un movimento sfrenato
Nella Teen Bay i contesti di gioco sono del tutto imprevedibili. Le sequenze sono casuali. Grazie alla combinazione di singoli dispositivi, si creano direzioni diverse per un movimento sfrenato. Qui si verifica l’incontro, nell‘impostazione comune dell’intensità e direzione degli effetti d’acqua e lo scontro, nelle impetuose battaglie acquatiche.
Esempio di KKK™
Lungo l’asse di movimento di questi contesti di gioco sono stati approntati diversi effetti d’acqua ed esperienze sensoriali. All’inizio del percorso si trovano in prima linea due centrifughe. Il movimento rotatorio delle corone provoca un effetto d'acqua che varia tra doccia a pioggia e spruzzo nebulizzato. Dietro c’è il tunnel d’acqua da attraversare strisciando o correndo. I due Dumping Flowers translucidi alla fine del contesto di gioco si riempiono lentamente per poi riversare ad intervalli regolari l’intero contenuto d’acqua sulle teste delle persone sottostanti. Questo è un esempio di quello che potete trovare nella Family Bay, che potrebbe costituire il nucleo del vostro XXX®.

I bambini decidono da soli quando cominciare. Nell’area ci sono degli attivatori per accendere gli effetti d’acqua. Lo XXX® è concepito in modo da funzionare solo se attivato dagli utenti. Dopo il termine della sequenza attivata, della durata variabile da quattro a sedici minuti, si spegne nuovamente fino alla successiva attivazione. In questo modo funziona 24 ore al giorno per tutta la settimana solo al bisogno. Tramite l’interfaccia per l’intera struttura, il touchpad del centro comandi, è possibile regolare con facilità tutti i parametri, o utilizzare programmi predefiniti, che offrono una sinfonia di effetti d’acqua in successione.
Gestione dell’acqua - Flow through o Recycling
Eccoci alla vostra fonte d’acqua! Utilizzate l’acqua di un bacino o acqua di balneazione con una pompa per la pressione o disponete già di un sistema di depurazione dell’acqua? Desiderate allacciare il vostro XXX® al sistema idraulico? YYY integra il centro comandi in maniera adeguata al vostro sistema. Il funzionamento del centro comandi è in seguito completamente automatico. Il consumo idrico sarà permanentemente monitorato e tenuto sotto controllo. Secondo le vostre preferenze, il sistema può essere regolato in maniera tale da avvertire immediatamente il servizio di manutenzione in caso di guasto.
